Dimensions and Theme in The Killers

On the eve of the grand economic crisis of the capitalistic world, the late 1920's is a turbulent period in the U. S. A. with violence and horror permeating the whole society underneath the surface of the temporary steadiness and prosperity. Among all the novels depicting the dark life then, The Killers is unanimously considered as the best for its superb technic and profound theme. The Killers offers a perfect example of a narrative that challenges and rewards the readers' perception of its structure. The structure of a Hemingway's story can usually be described, conventionally enough, with a set of scenes marked by a change in setting or by a change of characters. What is unconventional and so inscribes his fame as a modernist is that the scenes often are juxtaposed with little transition and less logic to effect or explain their sequence or rationale as a result of his prose style with its syntax linking sentences and simple causes without conjunction and subordination. This is not to say that the structures of the stories lack form or significant pattern, but they are coherent and connected in a sort of dimension which Hemingway said he learnt from cizanne's paintings. In The Killers, there are totally three main scenes: Henry's lunchroom, Hirsch's rooming house, and again Henry's lunchroom. The first scene opens with two strangers entering Henry's lunchroom, where George is waiting on Nick Adams at the counter. The strangers, Al and Max, try unsuccessfully to order from the diner menu, then settle for sandwiches, after which their small talk turns ugly. Al takes Sam the cook and Nick into the kitchen, gags and ties them up, and then he and Max reveal that they are waiting to kill the heavyweight prize fighter Ole Andreson when he comes to have dinner at six o' clock. At last, Ole Andreson does not arrive; the killers leave. In the second scene, Nick goes to Hirsch's rooming house to tell Ole about two men waiting to kill him and offers to tell the police; but Ole, lying on his bed and looking at the wall, says that he got in wrong; there is nothing to do and he is through running. In the third scene, Nick comes back to Henry's lunchroom and tells Sam and George his experience at Hirsch's rooming house. Sam won't listen to it and goes to the kitchen. The story ends as Nick wonders what Ole did, and George says; â€Å"Double-crossed somebody. That's what they kill them for. â€Å"I'm going to get out of this town,† Nick said. â€Å"Yes,† said George, â€Å"That's a good thing to do. † â€Å"I can't stand to think about him waiting in the room and knowing he is going to get it. It's too awful. † â€Å"Well,† said George, â€Å"you'd better not think about it. † In the first two scenes, the readers may get confused with the arrangement and disappointedly fail to wo rk out the theme, for despite all the impending violence and seemingly inevitable bloodshed, nothing happens: the killers do not kill and their victim still lies with his face to wall. The dimensions of the first two scenes seem to by default direct the readers' attention to the story's factual details: the two killers Al and Max are indistinguishable, dressed like twins or a â€Å"vaudeville team†; their sandwich orders are interchanged; they eat with their gloves on; they call George â€Å"bright boy† and suggests him going to see a movie; they get the lunchroom prepared for the killing; the nigger cook repeatedly complains and of George gives his careless remarks. However, no matter what image the readers have of the two dimensions, it will reflect the obvious symmetry between them: the action of the first scene is followed by the reaction of the second. And as the story moves on to the third dimension where the theme finally set in, the readers, with a reminiscent view, can figure out that all the details which seem irrelevant to the killing in the previous two dimensions, now, in re-perception, are closely knitted to serve the theme. The incident takes place when three men are in Henry's lunchroom: the cook Sam and the two waiters, George and Nick. Sam, from the very beginning, tries best to keep himself away from this killing: when Nick wants to go to inform Andreson, he advices Nick â€Å"to stay out of it†, â€Å"not to have anything to do with it at all. † Then Nick decides to go to Andreson in Hirsch's rooming house, he says: â€Å"Little boys always know what they want to do,† satirizing young men like Nick know nothing about possible danger in the unsafe society. Finally, Nick returns and gives his account of Ole Andreson's reaction, he won't even listen to it and shut himself in the kitchen. Having seen a lot of such violence of the society and got frightened by his own experience several minutes ago, Sam knows clearly the danger and darkness, and does not â€Å"want any more of that. † Thus he acts in a cowardly way to insure his own security. George also seems quite familiar with this sort of business: he becomes very suspicious of the two men when they mistake their own order and eat with their gloves on. As soon as the two killers mention Andreson, he knows their purpose of killing that boxer. Although he asks Nick to go to see Andreson, he does not care the whole thing much. When Nick asks him why the want to kill Andreson, he just make dismissive explanation â€Å"double-crossed somebody, that's what they kill them for. † For him, murdering is not a big event, since there are many â€Å"them† killed by â€Å"they† every day, he simply has already got used to it. Of the three men, Nick is the only one who shows great concern to the attempted killing. Innocent enough, he, in spite of Sam's warning, goes to inform the boxer. In Hirsch's rooming house, the boxer, although knowing himself in danger, lies on his bed helplessly and does not want to run. He tells Nick: â€Å"There ain't anything to do,† â€Å"I'm through with all that running around,† which gives Nick an even more terrible shock than that he is gagged and tied up by the two killers. So horrified Nick is, he decides to leave the town, â€Å"I can't stand to think about him waiting in the room and knowing he's going to get it. It ‘s too damned awful. † Thus, at this stage, the readers can see that the theme that Hemingway wants to express is not only to expose the violence and turbulence of society at that time, but in a more profound dimension, to reveal the shock and hurt of all these viciousness and wickedness to the innocent souls like Nick who will bear the image of that doomed boxer into his maturity. Therefore, although the title of the story is The Killers, the hero is actually the young man Nick, whose innocence in the setting of the cruelty of the killers, the cowardice of the cook, the indifference of George and the despair of Ole Andreson, refracts the instability and violence of the society at that disordered time. In The Killers, Hemingway, with his geometric design of the scenes, presents a story of simple plot but profound meaning, debunking the unsteadiness and turbulence of the apparently thriving society. Reading this story is just like viewing cizanne's landscape paintings: one has large room to perceive and appreciate its beauty and meaning in different dimensions.

The main components are: * Arts and Entertainment * Home-based Leisure * Countryside recreation * Catering * Heritage and visitor attractions * Sports and physical recreation Arts and Entertainment This is a huge industry that includes many sectors, they are: Arts The arts, apart from commercial cinema and theatre is a precarious industry where losses are easily incurred due to high costs and a very fragmented market. Sponsorship and grants play an important part in these. The public sector plays the most crucial part in funding arts. At the top is the Arts Council, which in 1999 gave out à ¯Ã‚ ¿Ã‚ ½188 million in grants. Much of this came from the National Lottery. Some of it was given out to Regional Arts Boards who then fund local projects. Much of the grant aid is given directly to organisations or individuals. The Arts Council is not the only national public sector organisation to be involved in the arts. One important group is The British Film Institute (BFI) which is responsible for supporting film makers, and the promotion of the British film industry. Local authorities play an important role as well. They employ arts development officers who promote the arts locally and will support local artists. Some larger authorities employ established artists in residence, who work in the area promoting their art and creating commissioned pieces. Local Authorities are major providers of galleries and performance space such as town hall stages or community theatres or even parish halls. In some cases, such as the Lyric Theatre at Hammersmith, London is a part owner of a commercial theatre. Voluntary sector groups also contribute to this sector. Entertainment Classified as entertainment are popular performing arts and spectacles. The enjoyment we get from them often depends on the environment we do them in. Competitive Sport-most people associate sport with the professional game. This means they are spectators, and it is the spectators money that underpins professional sport. Facilities were often run down to the extent that some were dangerous for crowds, e.g. Hillsborough 1989. Providers have found that profits increase when standards of customer care and corporate hospitality are improved. This has led to the building of a new generation of high-class stadia and race tracks and other facilities where the key services are: * Presentation of the core event * Merchandising * Catering * Safety and security Amateur sport is geared around participation and voluntary clubs. As land and buildings are expensive, many clubs use facilities provided by Local Authorities. Many outdoor sports clubs have their own facilities although there is an even greater number of players who use recreation grounds. There are about 78,000 pitches in the country. Thirty years ago amateur sport was largely centred on outdoor team games. Competitive sport means participation and developing skills and there is a sizeable industry for coaches and instructors. For most participants activity in the natural world is occasional, and more time will be spent in training in built facilities such as pools or climbing walls. Health and Fitness- there are now at least 2,500 private health and fitness clubs in the UK- it is a boom industry that many young people see as the exciting place to work in leisure. Private sector will increasingly dominate the market and eventually also manage and invest in public sector facilities on behalf of Local Authorities. Heritage and visitor attractions The heritage industry concerns buildings and materials that have historical value. Heritage was often about how the rich and famous had lived or what they had collected, or about culture(e.g. Shakespeare). Now it has expanded to include a much wider interpretation of historical value. Whether the attraction is a theme park or museum, the organisers will do their best to employ all the techniques of facility management to look after the customer and make the product interesting. In a theme park the excitement of the rides is a key feature and bringing new and more exciting rides brings in more customers. The difference between a visitor attraction and a heritage site is that the latter involves considerable work behind the scenes which the public never sees. Catering Leisure catering includes pubs and clubs, restaurants, cafes and takeaways. Its firmly placed in the private sector with some exceptions. A catering company provides a service according to the specification of the client and often under the clients name. Many leisure centres and theatres buy in service in this way. The catering sector has various components that make eating out an enjoyable experience: the provision of food and drink; entertainment, such as jazz at Pizza Express, games; and security in the form of door staff. Eating out is an increasingly important and available leisure experience. The fast-food business is booming with a growth rate of 30 per cent. This growth is at the expense of takeaways and cafes, and especially of pubs and clubs. Fast food tends to be run by chains that can make economies of scale by producing the same product in each outlet. Many breweries have followed this trend and operate chains like Harvester restaurants within their pubs. In contrast cafes, restaurants and takeaways tend to be run and owned by family businesses. Countryside recreation The countryside is the natural, national playground and one which is used by walkers, ramblers and those involved in more active outdoor activities. A survey in 1998 by the National Centre for Social Research showed that 1,427 million day visits to the countryside were made and a further 241 million to the coast. Like any leisure facility such usage will have an impact which in turn will lessen, and even threaten, the leisure experience itself. Organisations working in countryside recreation are therefore primarily stewards who maintain, manage and regulate the environment and also produce information and education. An important organisation funded by the Countryside Commission is the National Parks Authority. This is responsible for the regulation and interpretative service inside twelve National Parks, which are designated areas of significant natural beauty and wilderness. The Local Authority has a part to play in managing and regulating country parks. In many cases it also owns the land and will act as landlord to any tenants in the park, such as farmers. Home-based leisure This is cheap, easy and relaxing. The equipment most popularly used are: radios, videos, TV sets, gardening materials and home fitness machines is normally provided by the private sector. The main exceptions are the library lending services in the public sector. Except for broadcasting this component is retail based. Apoptosis And Inhibitor of apoptosis proteins Family: A   Background Apoptosis is an orchestrated biological cellular process that occurs in physiological and pathological conditions(1). It is essential for regulating development, homeostasis, and immune-system function in organisms(2). In mammalian cells, apoptosis is mediated by a family of cysteine proteases named caspases which are initially expressed in cells as inactive procaspase precursors and are activated by two pathways, the extrinsic ( or death receptor) and intrinsic (or mitochondrial) apoptotic pathways(1). The extrinsic pathway is activated by the binding of ligands such as Fas ligand (FasL) and tumour necrosis factor (TNF) to death receptors on the cell surface, FAS and the TNF receptor (TNFR), respectively, which leads to the formation of the death-induced signalling complex (DISC)(3)(4). DISC recruits caspase-8 and promotes the cascade of procaspase activation that follows(5). The intrinsic pathway is triggered by extracellular and intracellular stresses, such as high cytosolic [ca+2 ], hypoxia, severe oxidative stress, DNA damage(5), which results in the permeabilization of the outer mitochondrial membrane, the release of pro-apoptotic molecules such as cytochrome C and others into the cytoplasm(6), the formation of the apoptosome- a large protein complex that is made up of cytochrome C, apoptotic protease activating factor 1 (APAF1) and caspase-9 and caspase activation(7). On the other hand, cell death is also modified by other mitochondrial proteins such as apoptosis-inducing factor(AIF), second mitochondria- derived activator of caspase (Smac), direct IAP Binding protein with low PI (DIABLO)   and Omi/high temperature requirement protein A (Htr A2)(7). Smac/ DIABLO or Omi/HtrA2 induces cell death independently of caspase activation by counteracting inhibitor of apoptosis (IAP)- mediated caspase inhibition(7)(8)( Fig. 1). The upstream caspase for the intrinsic pathway is caspase 9, while that of the extrinsic pathway is caspase 8. The intrinsic and extrinsic pathways cleave the precursor forms of effector caspases, such ascaspase-3, caspase-6 and caspase-7(9). Activated effector caspases cleave many vital cellular proteins such as protein kinases, cytoskeletal proteins, DNA repair proteins and inhibitory subunits of endonucleases family and break up the nuclear scaffold and cytoskeleton(9). They also activate DNAase, that further degrade nuclear DNA(10), which together contribute to the typical morphological changes in apoptosis. Dysregulation of apoptosis has been implicated in numerous pathological conditions, including cancer(1). Besides, targeting the apoptotic pathways for cancer treatment is supported by several findings emphasizing the role of aberrant apoptosis in tumorigenesis and also resistance to anticancer treatment. Evasion from apoptosis is critical for tumor growth and a hallmark of cancer(11). One of the mechanisms by which evasion of apoptosis occurs is   disrupted balance of pro-apoptotic and anti-apoptotic proteins(1). A delicate balance between pro-apoptotic and anti-apoptotic mechanisms determines whether a cell death signal can activate the apoptotic program. It is not the absolute quantity but rather the ratio of these pro-and anti-apoptotic proteins that controls the regulation of cell death. In this balance, pro-apoptotic proteins activate apoptosis and anti-apoptotic proteins inhibit apoptosis(12)(13). Inhibitors of apoptosis protein (IAPs)   are important members of the anti-ap optotic family of proteins that can inhibit   caspase activation and play a key role in regulating of apoptosis in many species(1). Inhibitor of apoptosis proteins (IAPs): The inhibitor of apoptosis proteins are a group of structurally and functionally similar proteins that regulate programmed cell death, cytokinesis and signal transduction(14). The   IAP gene is 1.6 kb in size encoding a 31 kDa protein with a zinc finger-like motif. Many IAP family members have been identified in almost all species from viruses to mammals(15). They are characterized by the baculovirus IAP repeats (BIR) domain at the N- terminus, the name of which derives from the original discovery of these apoptosis suppressors in the genome of baculoviruses(16). The BIR domain contains approximately 70 amino acids. Although the number of BIR domains varies among IAP members, each BIR domain is made up of cysteine and histidine residues in a well-defined pattern (CX2CX16HX6C)(15). IAP acts as endogenous inhibitor of caspases by binding of their conserved BIR domains to the active sites of caspases in vitro and   vivo. IAPs inhibit caspases by promoting the degradation of active caspases, or by sequestering the caspases away from their substrates(17). When IAP family members are overexpressed, cancer cells no longer proceed to apoptosis and become increasingly resistant to standard chemo- and radiation therapies(18)(19). Many studies have established a circumstantial association between IAPs and cancer. Pathological overexpression of several IAP family members has been detected in several classes of human cancers(20)(21)(22). The eight IAPs identified in humans are cIAP1, cIAP2, NAIP, Survivin, XIAP, apollon, ILP-2 and livin(23). Interestingly, many data have shown that c-IAP1, c-IAP2 and XIAP are   broadly expressed in normal cells(24)(22). In normal tissues, IAPs could have some potential physiological roles, such as the regulation of the immune system(25), the response to cell damage(25), cell survival and differentiation(26). On the other hand, it has been proven in many studies that survivin, unlike other IAPs, is prominently expressed in vast majority of neoplasms but not in differentiated normal tissues(27). Survivin has been reported to be overexpressed in various cancers including breast and lung cancer, prostate, gastric, colon, bladder and esophageal carcinomas, osteosarcomas and lymphomas(28)(29). Overexpression of survivin was also found to be significantly associated with poor prognosis and decreased survivial rates in many cancers(30)(31). Survivin: Survivin (also Called IAP 4) is a protein with a crucial role in regulating both cell division and apoptosis. It is the smallest member of the IAP family(29). Survivin, a 16.5 kDa intracellular   protein of 142 amino acid, was discovered in 1997 by Ambrosini and colleagues(32). Structurally, survivin contains a single BIR domain. This domain is essential for its anti-apoptotic activity(33). However, instead of a ring finger domain (RING) near the C-terminus shared by others members of the IAPs, survivin contains a C-terminus alpha-helical coiled-coil (CC) domain which is thought to be important for its interaction with microtubules, hence its roles in cell cycle(34)(35) In normal tissues, survivin shows cell -cycle dependent expression during cell division. Its expression increases in G2/M phase and decreases rapidly in G1(29). The regulation of survivn expression and function is complex and can occur at various levels, including transcriptional regulation, post-translational modification, and protein stability regulation(27). it is regulated by a number of factors such as: NF-nB(36), insulin-like growth factor I/mTOR(37), Ras oncogene family(38), E2F, Sp1, TCF, and heat shock protein (Hsp) 90(39)(40).   Survivin is also regulated by p53 wild type. Additionally, post-transcriptional phosphorylation has been proven to play a   regulatory role in survivin activation(41). Biologic function of survivin Survivin as an inhibitor of apoptosis The mechanism by which survivin inhibits apoptosis is still controversial. Initially, survivin and other IAPs were postulated to inhibit apoptosis directly by interfering with the function of caspase-3, caspase- 7, and caspase-9(42).   In support of this model, it was shown that survivin can interact with Smac/DIABLO physically, thus placing survivin in a central position in the dynamic balance of proapoptotic and antiapoptotic factors(43). However, Structural analyses of survivin indicated later that any effect on caspase should be indirect, as it lacks the amino acid sequence that is essential in other IAPs for caspase binding. Also, the survivin gene is highly conserved in a wide range of organisms, and all of its orthologues are involved in mitotic regulation but not in cytoprotection(44).   Studies of cells from survivin-knockout mice have cast further doubt on the existence of a direct link between survivin and apoptosis(45). Later experiments indicated that Survivin inhibits active caspase-9 but not active caspase-3 and caspase-7. And, survivin mediated inhibition of caspase-9 requires interaction and cooperation with other molecules such as   HBXIP (hepatitis B X-interacting protein)(46) and   XIAP (X-linked inhibitor of apoptosis protein) which also known as inhibitor of apoptosis protein 3 (IAP3)(47) (Fig. 3). Survivin also provides cytoprotection to cells through the inhibition of the AIF pathway, which is known to induce caspase-independent DNA fragmentation(48). Survivin as a promotor of mitosis The cell- cycle dependent expression of survivin in normal tissues   supports strongly its role in cell division. During mitosis, survivin acts in a narrow time window at metaphase and anaphase. It is acting as an interphase between the centromere/central spindle and the chromosomal passenger complex (CPC)(49). CPC is a hetero-tetrameric complex which localizes to different sites at different times during mitosis, and   is composed of four components:   Aurora-B Kinase (enzymatic component), Borealin/Dasra, Survivin and inner centromere protein (INCENP)(50)(51). CPC is essential for proper chromosome segregation and cytokinesis(52). Inactivation of mammalian survivin -or its orthologues in lower organisms results in cytokinesis abnormalities, particularly spindle defects(53)(54) (Fig. 3)(55). Survivin facilitating angiogenesis In addition to its roles in apoptosis and mitosis, survivin   promotes angiogenesis. it is strongly expressed in endothelial cells (EC) during   the proliferative phase of angiogenesis(56)(57) and the antisense-mediated suppression of survivin   during angiogenesis stimulates vascular regression in vitro(58). Besides, exposure of cultured vascular EC to angiogenic factors such as VEGF and bFGF result in increasing survivin expression (both mRNA and protein)(59)(60). Survivin expression In normal physiological conditions, survivin is usually expressed in   embryonic lung and fetal organs in the developmental Stages(61). The protein is also detected in mature tissues with high proliferation potential such as thymus, placenta, CD34+ stem cells and basal colonic epithelial cells(61)(62)(63). However survivin seems to be selectively expressed in transformed cells and in most human cancers. Many studies have shown that survivin, unlike other IAPs, is prominently expressed in the vast majority of neoplasms but not in the differentiated normal tissue(27). Based on detection of protein by immunohistochemistry and mRNA by polymerase chain reaction techniques, overexpression of survivin has been reported in various human malignancies including lung cancer(64), breast cancer(65)(66); stomach(67)(68), esophagus(69), liver(70)(71), ovary cancers(72), brain(73) and hematological cancers(74). Additionally, the immunological responses which detected against survivin supports its specific up-regulation in malignant cells(75)(76). Survivin protein has also been shown to induce cytotoxic T-lymphocytes (CTL) response in   breast cancer, melanoma and chronic lymphatic leukemia patients(76). Survivin expression can be deregulated in cancer by several mechanisms, including amplification of the survivin locus on chromosome 17q25 (77), demethylation of survivin exons(78), increased promoter activity(79), and increased upstream signaling in the phosphatidylinositol 3-kinase or mitogen activated protein kinase pathways(80). Overall, increased survivin expression in several malignancies is associated with cancer survival or disease recurrence, and resistance to chemotherapy or radiotherapy. In a study of 275 patients with breast cancer demonstrated that survivin was a significant prognostic factor and predicted the outcome independent of patients age, tumor size and histologic grade(81). In the case of ovarian cancers, survivin expression was correlated with poor prognostic factors such as: high histologic  grade, mutant p53, and poor histologic type(81)(82). Also, previous studies demonstrated that survivin was expressed in benign brain and pituitary tumors. Although survivin   was also present in normal pituitary tissue,   the level of the gene expression was 6-fold higher in tumors than in normal pituitary tissue(83). In a study of 222   patients who underwent radical cystectomy, survivin was expressed in 64% of bladder tumors and 94% of malignant lymph nodes, but not in normal bladder specime ns and its expression correlated with disease recurrence and disease-specific mortality(84).   Also, increased survivin expression has been associated with an unfavorable survival or disease recurrence in colorectal cancer(85), particularly in stage II disease in esophageal cancer(86), hepatocellular carcinoma(87), lung cancer(88), glioma(89), leukemia(90), and other cancer types. A   study in oral cancer demonstrated that the extent of survivin expression was negatively correlated with the degree of differentiation(91). Additionally, survivin overexpression may be a predictive factor to determine response to chemotherapy and radiotherapy in patients with bladder cancer(92), breast cancer(93), multiple myeloma(94), lung cancer(95) and lymphoma(96)(97). On other hand, patients with lower survivin expression were more responsive to preoperative chemotherapy with 5-flourouracil and cisplatin in esophageal cancer(98). It is also reported that patients with lower survivin expression in pretreatment biopsies were more responsive to radiotherapies in rectal cancer(99). While Overexpression of survivin was associated with   resistance to a taxol-based   therapy for ovarian carcinomas(100). In addition to full-length transcript (survivin (wild type)), five splice variants, which result from splicing of survivin BIRC5 gene pre-messenger RNA (mRNA), have been described: survivin-ΔEx3, survivin-3B, survivin-2ß, survivin2ÃŽ ± and survivin 3ÃŽ ± with different structure and function(101)(102)(103). Previous studies showed that an imbalance in the alternative transcript ratios may affect the cell to be resistant or sensitive to apoptosis(104). This alternative splicing of Survivin has been shown to have correlation with disease activity in various patient studies. For example, studies showed that Survivin-ΔEx3 and survivin-3B were found to be highest in tumors with advanced histological grade and were associated with poor prognosis(105)(106). On other hand, the expression of survivin-2ß was significantly higher in small tumor size and was inversely associated with axillary node positive carcinomas(106). Besides different splicing forms, immunohistochemical studies have demonstrated that survivin also localized   in distinct nuclear and cytoplasmic subcellular pools. Cytosolic Survivin is believed to act as apoptotic suppressor while nuclear Survivin is postulated to regulate cell division(29). There are conflicting data of pathological significance of nuclear Survivin.   Some Splicing studies showed that nuclear staining of survivin is associated with favorable prognosis(107), while others showed Its expression in the nuclei of tumor cells appears to be associated with unfavorable clinical outcomes(108)(109). Also, the cellular localization of Survivin isoforms   differs. while survivin-2ß   and Survivin 2a are localized in both nuclear and cytoplasmic compartments, survivin-ΔEx3 is localized in both mitochondria and nucleus(110). Additionally, Methylation and Phosphorylation are critical requirements for survivin function. Several observations show that survivin is unmethylated in cancer but may be selectively methylate  d in normal tissues with individual variations(111)(112). Methylation may play an important role in the p53 mediated suppression of survivin(113). Another critical requirement for survivin function is the phosphorylation on Thr34(114) Treatment approaches: Due to important role of Survivin in tumor cell division, apoptosis, chemo resistance and survival, survivin represents a unique target for biologic therapy in many human malignancies. Several novel experimental therapeutic strategies have been developed to block the expression or function of Survivin in tumour cells. These include immunotherapeutic approaches to induce immune response against Survivin, small molecule inhibitors/antagonists of   survivin function, and nucleic acid based approaches which interfere with Survivin gene expression(115)   such as antisense oligonucleotides (ASOs), ribozymes and small interfering RNAs (siRNAs)(116). Also, Vaccine approaches such as dendritic cell based (DC) vaccines, DNA vaccines(117), peptide vaccines for Survivin have also been evaluated in preclinical or clinical studies. Survivin ASOs were first used against malignant melanoma cell lines. Transfection with the ASOs triggered spontaneous apoptosis linked to decreased endogenous survivin expression(118) . Treatment with LY2181308, a specific inhibitor of Survivin mRNA which has already entered the phase 1 trial(119). YM-155 is a novel small-molecule survivin suppressant which inhibits survivin mRNA transcription and protein expression in p53-deficient cancer cells in vitro(120). YM155 has also shown to be effective in vivo models of prostate, pancreatic, and lung cancer(120)(121). Ribozyme mediated approaches have also been evaluated for inhibition of Survivin expression. In addition, the number of the users is highly increas- ing through the time and as result the related databases start overloading. Furthermore, despite the fact that data indepence would be ideal for im- plemeting lithe applications developers abandoned this idea in order to avoid expensive queries. Hence, Michael Armbrust et al.[1] implemented a new declarative language called PIQL, a scale independent language. A large number of frameworks have already appeared in order to assist developers to create modern web applications. However, this plethora of websites with millions of users led to database failures due to lack of request managing. As a result, there was a demand on implementation of a new system that will control all these requests and provide efficient results to users. A few methods have introduced and one of the most popular is NoSQL. Despite the fact that NoSQL provided a high level interface, data indepen- dence created scalability problems since a large number of queries took a lot of time. This led to to several issues like performance failing and user disatisfaction as well. In order to avoid this bad situation scientists hand coded key/value implementations. On the one hand, this provided the de- sirable scalability but, on the other hand is was not easy enough for the developers to write that kind of code to parallelize their queries so as to fi achieve high scalability. Another significant issue is time consuming functions rewrites. Now, once we talked about several problems occured by queries in the next section we will discuss about PIQL. More specifically, we will present this method and give a brief summary of the implementation. In the rest of the document we will discuss about the performance of the previous imple- mentation. What is PIQL? In this section we will discuss and analyze the PIQL (PerformanceInsightful Query Language) model. One important advantage of PIQL is that intro- duced the notion of scale independency. More specifically, the model pre- serves the logical data independence. The most significant about data this technique is that performace maintains not only on small datasets but also in large as well. For this reason this is called success-tolerant since the success is for every large dataset. But why PIQL is successful? The answer is on the limitation on key/value store operations. As we previously mentioned, one goal of PIQL is to avoid issues when the database gets larger. PIQL uses static analysis in order to fi the correct number of operation in every step of the execution. Before we move to the next step of the analysis of the methodology we should mention the four queries classes. The fi one is called constant since the processing time is constant. The second one is the bounded class. More specifically this class refers to bounded data when the site becomes more popular. For instance, in case of Facebook every user has a limit of 5000 friends. The third class is called sub-linear or linear and is referring to queries that become more successfull when the data increase linearly. The last one is Super-linear where intermediate calculations are necessary for the queries. Now, once we mentioned all the necessary theoritical parts of PIQL we will discuss on its structure. Every server is directly connected with a Distributed Key/Value Store. Hence, this methodology maintains the scalability and the response time is now predictable. A significant drawback of this technique is that a specific key/value store is required so as to maintain data locality. On the other hand, this method is non-blocking and according to Chen et al.[2] can reduce memory latency. Another important benefit of PIQL is that extends the cardinality con- straint of regular direction to diff ent directions as well. More specifi , these cardinalities provide several information on its relationships. For in- stance, a Facebook user should have no more than 5000 friends. This is a very significant information since selecting the wrong number for limita- tions can lead back to the previous problems. Thinking again the Facebook limitations for the maximum number of friends on Facebook, according to Brandtzg et al.[3] a significant issue that occurs is the lack of privacy. Hence, the limitations are not only important for the performance but also for the user protection as well. In addition, the same person can create a new profit for free and add his new friends there. As a result, 5000 friends is not actually a limitation for a user and is provided in terms of privacy and performance. According to Michael Armbrust et al.[1] their algorithm for scale inde- pendent optim ization contains two phases. The fi one is reffering to stop operator insertion. In order to maintain scalability, the algorithm starts by fi a linear join ordering on the query parser. Depsite the fact that stop operator is already contained due to LIMIT which in contained in the reg- ular query, scientists have introduced data-stop operators which are pushed in lower levels in order to preserve the initial rules without the demand of  restart the whole system. Next, after fiphase 1 the second step which is called remote op- erator matching. As we previously mentioned we should ensure scalabiliy. Hence, the intermediate results are bounded. But how all these logical op- erators are mapped on remote operators? For Index Scan, that means that maximum one attribute can be affected by predicates. Analysis of The Kids Are All Right by Susan Faludi Kids are crawling around in the dirt, screaming, and have not yet had their diapers changed because the day care provider seems to be in a trance watching the latest episode of the Montel Williams show. One of the workers strikes a child because she won’t stop crying about how hungry she is. The other worker just sits in her chair drinking Jack Daniels with a little Coke mixed in. Not all is well at the Wee World Child Center. But is this the impression that the public perceives of our daycare system in America?   Ã‚  Ã‚  Ã‚  Ã‚  Well, most people would say that this is how only a few daycares are run. But many people would still state that kids who have not been in daycare have a better chance at a more enjoyable life than those who have. Susan Faludi, who frequently writes about women’s issues and is the author of Backlash: The Undeclared War Against American Women, promotes daycare as an enhancement in a child’s life. In her essay, The Kids Are All Right, she claims that kids who attend daycare are more social, experimental, self-assured, cooperative and creative. Faludi’s argument is convincing because she provides solid authoritative sources, gives personal experiences of other girls who have been in day care, and refutes other researchers claims. Susan Faludi dives right into her argument and hits us with an informative source. Faludi cites Alison Clarke-Stewart, a professor of social ecology at the University California at Irvine, who found that social and intellectual development of children in day care was six to nine months ahead of children who stayed at home. This source is reliable because the author of the statement is an expert in the field of social ecology. Therefore this is an opinionative informative source because the researcher could be biased toward one side of the argument or the other. This matters to Faludi because audience could question the reliability of the source.   Ã‚  Ã‚  Ã‚  Ã‚  Susan Faludi also cites personal experience in the form of interviews done by Delores Gold and David Andres in paragraph number two. The interviews of the girls provide not only data on childcare accountability, but also serve to put a personal and more intimate effect on the argument. The interviews have a great effect on the reader because they are grounded in reality and have been conducted by experienced researchers.

Teaching Philosophy :: Education Learning School Essays

Teaching Philosophy Teaching is learning twice over. I believe that learning can be done in many different capacities; however, it takes a strong blend of certain personal characteristics and instructional techniques to become an effective teacher. In a perfect world, I could stand in front of a gathering of physical education students and teach them successfully how to dribble a basketball or throw a baseball, but it is much more difficult than that. Students are as different as snowflakes, each with many characteristics different than the next. To use the analogy of a key and lock, the key being the presentation of the material and the lock being the students, I feel that many teachers try to use the same key on 25 different locks. It is up to the teacher do away with the â€Å"here is the material†¦learn it† method of teaching and start considering the differences between one student’s ability to learn to the next. I believe in having a strong relationship with my students. I want them to be in a classroom where they know someone cares about them and that the teacher is not just earning a paycheck. By demonstrating a certain level of compassion for your students, they feel more comfortable with you as a teacher. By nurturing their open minds, a teacher can be successful. It is important to let the students ask questions and let them think for themselves. At a young age, children are full of imagination and ideas, and too often teachers take their intuitive instinct away from them by not letting them share their ideas or punishing them when they make a mistake out of the exploration of their imagination. I believe that setting goals has got me where I am today and that the concept of goal setting should be instructed and reinforced. Without goals students are apt to stray from little educational endeavors that end up being the little holes that sunk the big ship. In high school, I was a student who was at the top of the class in subjects that I enjoyed and mediocre in subjects that I did not find appealing.

12th Night - Orsino Essay -- essays research papers

  Ã‚  Ã‚  Ã‚  Ã‚  William Shakespeare’s Twelfth Night depicts the trials and faults of several characters’ loves. There are many downfalls and unrequited loves, and the story basically ends up in a confusing love triangle. He especially shows the many quirks of Orsino in his quest for winning the true love of Olivia. In this play, the reader can easily understand the many mistakes that Orsino makes in love.   Ã‚  Ã‚  Ã‚  Ã‚  For a majority of the play, Orsino is very oblivious to the fact that Cesario is actually a woman. Viola, disguised as Cesario, makes many comments to him that could possibly lead him to finding out her secret. For example, when Orsino asks what kind of woman Cesario loves, she replies â€Å"Of your complexion† (2.4.27). Orsino does not catch this, but describes that one should love a woman younger than himself. â€Å"For women are roses, whose fair flower Being once displayed, doth fall that very hour† (2.4.40-41). This is most likely a positive point for Viola, being that she is clearly younger than Orsino, and once the disguises are taken away, he will realize that he can love her. Orsino 1 actually describes a platonic love between himself and Cesario. This is a hint to the reader that the unveiling of Viola could, in fact, lead to a true love. For instance, Orsino tells Cesario â€Å"If ever thou shalt love; in the sweet pangs of it remember me† (2.4.13-14). This is almost ironic, and foreshadows the follies yet to come including the growin...

Chaos theory

What exactly is the chaos theory? Some believe the chaos theory is one of the many theories that will be recognized in the centuries to come. The chaos theory embodies many conditions of science, such as physics, engineering, economics, philosophy, mathematics, music, and even psychology. The chaos theory is only beginning. The chaos theory is a theory used in different categories of science that a seemingly possible phenomena has an underlying meaning. When was chaos first discovered? Edward Lorenz was the first true experimenter in chaos, he was a meteorologist.In 1960 Edward Lorenz was working on a weather prediction problem, he ad a computer set up to model the weather with twelve equations. His computer program did not predict the weather, but theoretically predicted what the weather might be. In 1961 Edward Lorenz wanted to see a specific sequence again, to save time he began in the middle of the sequence. He entered his printout number and let it run. An hour later the sequenc e had changed differently. The pattern had diverged, ending up being extremely different. His computer had saved the numbers to a six decimal place, he printed it out for three decimal places to save paper.The original sequence was 0. 506127 he had it as 0. 506. Lorenz's experiment: he ditterence between the sta rting values ot these curves is only . 000127. (Ian Stewart, Does God Play Dice? The Mathematics of Chaos, pg. 141) This is how the butterfly effect became, because of the number of differences of the two curves starting points was that of a butterfly's wings flapping. Unpredictability is one of the most important element is a complicated system. Lorenz calls this unpredictability â€Å"sensitivity to initial conditions,† which is also known to be the butterfly effect.This idea means with a non-linear, complex systems, starting conditions will effect in extremely dissimilar outputs. The effect of the utterfly's movements, to predict the weather. An example is if a but terfly flaps it's wings in Tokyo, it could predict a storm in Texas in several weeks time. The dependance on initial conditions is extreme. There is a rule for complicated systems that one cannot create a model that will predict outcomes accurately. The idea initial conditions on sensitive dependance mathematical roots are powerful.If you have a circle with the points XO and Xl , this represents the starting value for a variable. â€Å"We assume that the difference between there two numbers is represented by the distance between the points on the circle, given by the ariable d. In 1963 he published a paper describing his discovery and the unpredictability of the weather. This paper also included key information about the types of equations and what caused this behaviour. Since he was not a mathematician or a physicist he wasn't cknowledged for his discoveries until year later, when there had already been rediscovered by others. Lorenz had to wait for someone to discover him, his discovery was revolutionary. Another example of sensitive dependance of initial conditions is flipping a coin. There are two variables.One is how fast it is flipping, and the other is how fast it will hit the ground? Apparently, it should be likely to control how might the coin end up. In practice, it is impossible to control exactly how fast the coin flips and how high. There are similar problems like this in ecology. This occurs with the prediction of biological population. If the population rises continually, but with predators and limited food supply the equation is incorrect. next year's population = r * this year's population * (1 – this year's population) Benoit Mandelbrot was a mathematician working at 18M, he was studying self- similarity.One ot the areas ne studying was cotton prize tluctuation. He tried many times to analyze the data of the price for cotton, but the data did not go with the normal distribution. He decided to collect data from. Mandelbrot eventually gave up, until he decided to gather all the information dating back to 1900. He IBM's computer and found a surprising fact: The numbers that produced aberrations from the point of view of normal distribution produced symmetry from the point of view of scaling. Each particular price change was random and changes unpredictable.But the sequence of changes was independent on scale: curves for daily price changes a nd monthly price changes matched perfectly. The heart has a chaotic pattern. The time between the beats is not constant. It depends on how much actions a person is doing and there also among other things.

Fashion of the Roaring Twenties and The Sixties Fashion is the style prevalent at a given time. It usually refers to costume or clothing style. Everybody has to wear clothes, making fashion a part of everyday life. The way someone dresses says a lot about his or her personality, age, culture and experience. At times of economic or social change, fashion often changed. The 1920s and the 1960s are big eras were economic and social change were happening. They are both largely known for their fashion.The 1920s was also known as the Roaring Twenties due to the period’s social, artistic, and cultural energy. The twenties were right after the end of World War One and right before the Great Depression. The era was notable for inventions and discoveries, industrial growth, increased consumer demand and significant changes in lifestyle (â€Å"roaring twenties†). During the twenties, the economy of the United States evolved from a wartime economy to a peacetime economy. The Eighte enth Amendment prohibited the manufacture, sale, import and export of alcohol in attempt to help the social problems.Passing the nineteenth amendment gave women the political equality they had been fighting for. The twenties were also known as the Jazz Age because jazz music grew in popularity. â€Å"During the 1920s jazz music flourished, the flapper redefined modern womanhood, Art Deco peaked, and the Wall Street Crash of 1929 ended the era, as the Great Depression set in† (â€Å"roaring twenties†). The Roaring Twenties were trying to break from traditions of the Victorian way of life. Since the 1920s was a time of celebration, there were many fads.Young women’s fashion of the 1920s was both a trend and a social statement. They were labeled ‘flappers’ by the older generations. â€Å"Flapper† was a popular slang for a very young prostitute or a lively mid-teenage girl. â€Å"The image of flappers were young women who went to jazz clubs at n ight where they danced provocatively, smoked cigarettes through long holders, and dated freely, perhaps indiscriminately. They rode bicycles, drove cars, and openly drank alcohol, a defiant act in the American period of Prohibition† (â€Å"flapper†). They were seen as rude and self-assertive for their behavior. The ‘new’ woman was less invested in social service than the Progressive generations, and in tune with the capitalistic spirit of the era, she was eager to compete and to find personal fulfillment† (â€Å"roaring twenties†). Flappers as a social group were separate from other 1920s groups; their behavior was bizarre at the time. They challenged women’s traditional public roles, supporting voting and women’s rights. Some flappers weren’t into the politics. â€Å"Older suffragettes, who fought for the right for women to vote, viewed flappers as vapid and in some ways unworthy of the enfranchisement they had worked so hard to win† (â€Å"flapper†).In addition to their strange behavior, they were known for their style. Flapper style made girls look young and boyish. They had a chin-length bob hairstyle and wore straight waist dresses with a hemline above the knee. The risen hemline allowed flashing of the legs when a girl danced. They also removed the corset from female fashion. Until the 1920s, cosmetics were not accepted in American society because of its association with prostitution but flappers made cosmetics popular. Another popular era for fashion was the 1960s, more commonly called The Sixties. The sixties was a movement escaping from the conservative w ays of the fifties and eventually resulted in revolutionary ways of thinking and real changes in the culture of American life (Goodwin).The Civil Rights Movement played a major role of changes in society in the 1960s, starting with Martin Luther King Jr. In 1961, John F. Kennedy was elected president, becoming the youngest president to ever hold office. The Vietnam War was a major event in the 1960s that absorbed a lot of national attention. The draft took place followed by anti-war outlook. The National Organization of Women questioned the unequal treatment of women in the society. The hippie movement arose as a result of young people not content with the generation ahead of them. The sixties was a decade that broke many fashion traditions, mirroring social movements during the period† (â€Å"fashion†). The sixties were the age of youth, who wanted change. The changes they were looking for affected education, values, lifestyles, laws and entertainment (Goodwin). There were so many young people in the sixties due to the post-war baby boom. The children had grown up and were becoming teenagers and young adults. These youths changed the fashion, the fads, and the politics of the decade (Goodwin).They dressed to convey rebellion. â€Å"The sixties began a decade which may well be recorded as one of the most fashion-conscious periods in recent history, challenging the 1890s and the 1920s† (â€Å"1960s fashion†). The hippie movement arose during the mid-1960s. Hippies didn’t focus on what others thought, they believed in ‘equality for all’. Hippie characteristics comprised of listening to rock, accepting sexual revolution and the use of drugs such as marijuana and LSD to explore alternative states of consciousness (â€Å"hippie†).Hippies were pacifists, the belief that any violence is unjustifiable, under any circumstances, and that all arguments should be settled by peaceful meanings. They participated civil ri ghts marches and anti-Vietnam War protests. â€Å"The late 1960 produced a style categorized of people whom promoted sexual liberation and favored a type of politics reflecting ‘peace, love and freedom’† (â€Å"fashion†). By 1965, hippies had become an established social group in the United States. By 1968, the hippie look was in style. Hippie’s way of dress and grooming was one way of expressing their thought of independence.Both genders wore bell-bottomed jeans, tie-dyed shirts, headbands and sandals. Women often went barefoot and braless. Teenage girls often wore fringed buckskin vests, flowing long dress, Mexican peasant blouses, gypsy-style skirts, scarves, and bangles (â€Å"fashion†). Fabrics frequently consisted of animal or paisley prints. Both men and women grew their hair out long, including men’s facial hair. Further trends included love beads, peace signs, body piercing and tattoo body art. Hippies wore clothes that they f elt communicated themselves and their individualism. Hippies repelled the post war ugliness in the world and turned the attention of fashion stalwarts towards as much natural beauty as possible† (Borade). The 1920s and the 1960s style of fashion appears to be very diverse. When observing each era’s attire they portray different looks. In the twenties, they wore dark colors opposed to the sixties where they wore very bright colors and patterns. Flappers wore tight, short dresses while hippies wore loose, floor length dresses. Flappers popularized make-up, wearing very drastic blush and lipstick. Hippies wanted to be natural so they didn’t wear any make-up.Although they seemed completely different, their motives behind their eccentric fashion were the same. They were both rebelling against the norm and trying to prove their independence. Fashion is a way of expressing personality and independence. Every person has their own sense of style and they show that through the way they dress. Everyday people get dressed, choosing clothes that they like and illustrate them best. Clothes are talked about, thought about and worn daily. Each era has a different fashion that shows up in clothes, attitude, personality and behavior.

The cell phone has completely changed the way that the world lives. † We live in a world of different technologies, but the cell phone seems to have the biggest impact on peoples’ life today. We use newer cell phones and updates daily. Cell phones have changes drastically since the very first cell phone was invented in 1983. Although some people think cell phones updates have not affected them, it honestly has by having different prices of the cell phone, having new appearances’ of the cell phone, the way people communicate, and new updates for different apps.The cost of a cell phone has definitely changed over the years, for instance Retro Brick said, â€Å"the price was some $3,995 in 1983 dollars. † Back in 1983, $3,995 was a large amount of money. Comparing the amount in 1983 to 2013, $3,995 is like probably thousands and thousands of dollars more than that in 1983. Today in America to buy a cell phone, you need a c ontract with a cellular provider for example AT&T, Sprint, and Verizon Wireless.Lets’ say the buyer wants to buy the new IPhone 5, they would have to buy the data plan that has internet, texting, and calling; the costumer has a choice of unlimited calling and texting or having a certain amount minutes for calling and a certain amount of text messages you can send. The price of the IPhone 5 64GB alone is $400. With a contract the IPhone 5 64GB comes out to about $200. The appearance of the cell phone has changed completely. In 1983 the phone was super big, as NBC describes it, â€Å"the brick†.It would only allow thirty minute calls and took up to eight hours for it to charge. It was gray and had a large keypad. The only thing you could operate was have a call for thirty minutes. You could only save a few numbers. Of course if you can only have phone calls, which means you could not take pictures. The IPhone 5 is very thin and is light in weight. You have a choice betwe en white or black phone. It only takes up to about forty-five minutes for it to fully charge.It is touch screen, you can take pictures, listen to music, save up to hundreds of phone numbers, and you can send text messages. You can also get onto the internet. Since the first cell phone only allowed one thirty minute conversation, you usually had to talk to people in person more and would have to write letter instead of just sending text message. They would usually have to set up meetings. Which today you can have a ten hour phone call if you wanted too and you can send text messages if you need to tell them something at the exact moment, or they have FaceTime meetings, o it really changed how people communicate in today’s society. When the first cell phone came out of course it did not have any apps for it if all you could was call and receive calls. Which today there are over thousands of apps to get. Many people buy or download new apps on a daily basis. There are different types of apps which are social media, games, news, books, business, education, fitness, medical, lifestyle, music sports travel, utilities, and weather.The app that mostly all teens and adults have is Facebook, Twitter, and Instagram. It’s usually teens that have games and entertainment apps on their phones. Adults usually have books, weather, education, and all the other adult categories of apps. There are always new updates for all of these apps. For instance Twitter just had an update and the last update was a couple months ago, meaning usually apps have a new update with new features every couple of months, or even sooner or later than that.It usually depends on the app producer. There will always be new appearances’ of cell phones and different prices.
